Research at a glance

Ipamorelin is a synthetic pentapeptide characterised in the literature as a selective agonist of the growth hormone secretagogue receptor 1a (ghrelin receptor). Preclinical work has studied it chiefly in animal models of feeding, reproductive endocrinology, and gastrointestinal motility, alongside analytical and clinical investigations in humans. The evidence base is predominantly animal and in vitro, with a small number of controlled human studies; broader controlled human data remain limited.

  • In a cichlid fish model (Oreochromis mossambicus), administration of ipamorelin acetate over 21 days was associated with dose-dependent increases in food intake and in spermatocyte and spermatid counts, alongside elevated androgen receptor expression and serum luteinizing hormone and 11-ketotestosterone, within a study of the hypothalamic-pituitary-testicular axis.[1]

  • In a randomized, double-blind, placebo-controlled phase 2 proof-of-concept trial in adults undergoing bowel resection, intravenous ipamorelin was evaluated for safety and for time to tolerance of a standardized solid meal in the setting of postoperative ileus.[2]

  • In a human urinary study following nasal administration, ipamorelin was reported to metabolise extensively and to be excreted as a set of parent compound and metabolites, with the Ipamorelin (1-4) free acid identified by nano-liquid chromatography–high resolution mass spectrometry in the context of anti-doping analysis.[3]
